the state constitutions commerce clause gives the legislature the power to write laws regarding the regulation of commerce. The chapter of florida law I pasted explains all the commerce regulating power. The legislature can then regulate that commerce to either require cruise lines to verify vaccine status or prohibit cruise lines from verifying.

Desantis just want to spread the theory of Don that the virus is a hoax. He wants to use his power to push it in industries that are more interested in building business. Getting vaccines for all employees and demanding them from those who want to go, makes sense for them.

Also, there are thousands of cases of people who had COVID once, and then contracted it again.
