I would say it doesn't necessarily have to be a member of Congress who is a party 'leader'. In the Republicans case right now I'd say it's pretty much a rudderless ship as far as leadership. While Boehner and McConnell are the highest ranking Republicans in office I wouldn't really call either of them "leaders".
